You can get a x-eng brake to fit on a transfer case with an overdrive fitted, surprised you haven't got that version.

I fitted an Ashcrofts Transfer box to a 110V8 at the weekend. At the same time I fitted an X-brake. The vehicle also has an O/D and there were no problems at all with fitting the brake.Les.
Wouldn't have thought this a problem on defender as the calliper fits above the disc rather than to the left side in the case of Disco and RRC. Fitted one at the weekend was fun, Was interesting as the instruction seem to be for the original design but we manage it anyway.
As above - the inlet pipe on the PTO pump caused a little bit of grief, but nothing serious. The caliper on mine sits over at a slight angle towards the offside, yet the instructions hint that it should sit on top of the disc......unless I put the backplate on wrong ..........I fitted the backplate with the disc guard tab parallel to the ground, I could rotate the plate anti-clockwise, and then the caliper would sit on top the disc, but then the guard tab would no longer be guarding the disc :-.
all is well..........just need to find a 45deg slope to try it out :D
